Welcome to SMARTrial shiny app. This application can be used to explore data from the SMARTrial at the German Cancer Research Center (DKFZ) and the University Hospital Heidelberg. In the SMARTrial cohort primary cancer cells from patients with leukemia or lymphoma and treatment indication are tested for their in-vitro drug sensitivity. In the Validation cohort, only AML patients were included.

Select samples for analysis

In this tab, you can subset the SMARTrial dataset based on the below criteria. The subsetted dataset will be used for the analyses in other panels.

Patient data from 2 patients were excluded due to high technical noise (SD of negative controls > 0.3). Further, for 9 patients, in vivo response was not evaluable, these were excluded here.

Information of selected patient samples

Heatmap plot of the normalized counts on screen plates

This tab shows the viability distribution on the 384-well assay plates. It is mainly used for quality control purposes.

Dose-response curves for specified patient samples

This tab allows you to inspect the sensitivity on single drugs or drug groups. You can either select a single drug or a group of drugs by their target or pathway annotation.

You can also color the patients by those features using the Color by option.

Boxplot visualization

This tab allows you to visualize the association between drug response and a selected patient characteristic, such as in vivo response using a boxplot

The associations P-values from ANOVA test will also be indicated in the plot title.

Ranking drug effect for a selected patient

In this tab, the response of the selected patient to all drugs is compared to the median response across all patients. These responses are then ranked to show the drug with the strongest relative response in the in-vitro sensitivity assay. Both the drugs and the patients can be subset here.
